If a single user toilet room is added to a second story of a building that does not require vertical access e.g. an elevator, does the single user toilet room toilet need to be made accessible?

A hotel room is occupied by a guest with a service animal. The hotel guest provides a mat for the animal to urinate on in the room. The mat has been allowed to overflow. The hotel manager wants to know if the hotel can do anything about the situation.

The single user toilet room complies with the 1991 ADA Standards for Accessible Design, and the sink overlaps the space required at a toilet. Does the single user toilet room have to be renovated so that it complies with the 2010 ADA Standards for Accessible Design?
